Any chance you could ask your wife's grandfather where he got it? It looks real to me given the folds and imperfections around the edges, but it's still in suspiciously good condition; and fake Chinese banknotes are also often coming out of China.

Yes, like Oklahoman said, the big grading companies (PMG for banknotes) are generally trustworthy, although it might not be worth going to basement companies even if they offer services at a lower rate.
I don't have any graded notes myself; I bought a few in WAC (a French grading company) certified holders, but cut them open as soon as I got them.
I would not call a note with a catalog value of about $12-20 a "valuable" note. This is a 1949 note, P-853 in the catalog. The condition seems to be VG-FINE. If it was obtained in this century, it could very well be counterfeit. The usage marks may or may not be real.
A couple of Chinese sellers are counterfeiting the 1949-1960 notes and selling them to suckers on ebay by the hundreds. I have about 20 of these counterfeits and I buy them on ebay if I can get them for 25¢ or less. I once had a set of 60 purchased for $6 but I traded it away.
